EVs isolated from TIS & control cells were immunoblotted to validate the protein expression of annexin V, Na/K ATPase & alpha -tubulin from the MS data. CD63 & HSP70 were used as loading controls. Densitometry was completed using Image J software (Fiji). All values are expressed as the mean of three independent experiments±s.e.m. The limit of ±1 skewness was set as normal distribution. ![Western Blot: Survivin Antibody (60.11) - BSA Free [NB500-238] - CD133+ cells are more resistant to chemotherapeutic agents. STA-ET-8.2 cells were FACS-sorted & CD133+ & CD133- fractions treated with increasing concentrations of (A) Doxorubicin (Doxo), (B) Etoposide (Etop), (C) Vincristine (Vinc) or (D) a combination of all three drugs at high (Doxo 10 μM; Etop 10 ug/ml; Vinc 100 ng/ml), medium (Doxo 2.5 μM; 20 Etop 2.5 ug/ml; Vinc 25 ng/ml) & low (Doxo 0.5 μM; Etop 0.5 ug/ml; Vinc5 ng/ml) concentrations. SYCP3 is a protein essential for synapsis of homologous chromosomes & gamma H2AX localizes to double-strand breaks & XY bodies during meiosis. A total of 200 spreads were counted in testis cell preparations from WT & GCKO mice at P22. (B) Chi square analysis shows GCKO testes contained significantly higher numbers of germ cell spreads at the leptotene & zygotene stages of meiosis I & fewer spreads at pachytene, diplotene & metaphase I stages (P<0.001), suggesting that the loss of Dicer1 lead to disruptions in progression through meiosis I. Immunomagnetic anti-CD45 depletion was performed on the obtained PBMCs & the remaining cells were washed, cytospunned onto the slides. Slides were stained for PSMA, EpCAM, sLex, & CXCR4 using the protocol as described in Figure S1. MDA, PC3, & KG1 cells were simultaneously stained as a control for the following markers: PSMA= Magenta, EpCAM= Yellow, HECA-452= Green, CXCR4= Red. All prostate CTCs expressed CXCR4, while, sLex expression was variable. The analysis of sLex intensity is shown in Figure 4.
